துரைசாமி, ஜானகி தம்பதியருக்கு மகளாக சிறீவைகுந்தம் அனும் ஊரில் பிறந்தார்S. சிறுவயது முதலே மேடைநாடகத்தில் ஆர்வம் காட்டினார். பின்நாளில் அவரது குடும்பம் மதுரையில் வாழ்ந்தனர். அங்கே அவர் கருநாடக சங்கீதமும், நாட்டியமும் பயின்றார். அவரது பெற்றோர் பலவாறு இவருக்கு ஒப்பனையிட்டு நாடக நிருவனங்களில் இவரது புகைப்படங்களை அனுப்பி வாய்ப்புகளைத் தேடினர். இவ்வாய்ப்பே இவர் சிறுவயது முதல் மேடை நாடகங்களில் பங்குகொள்ள உதவியது. வளர்ந்த பின்னர் இவர் [[எம். கே. தியாகராஜ பாகவதர்]], [[கே. பி. சுந்தராம்பாள்]] மற்றும் [[டி. ஆர். மகாலிங்கம்]] போன்ற முக்கிய நட்சத்திரங்களுடன் நடிக்துப் புகழ் பெற உதவியது. Letchumanan Chettiar or Lena Chettiar of Krishna Pictures who has seen the drama wanted to picturise the drama and requested [[K. Subrahmanyam]] to direct that film. Lena Chettiar and K. Subramaniam watched the drama together. K. Subramanyam suggested to retain the same artists who acted in drama to be in the cinema too and Lena Chettiar agreed to this. Through [[Pavalakkodi]], S. D. Subbulakshmi entered into Tamil Cinema in 1934. She became a famous heroine in the 1930s till mid-1940s, then she switched to mother roles and supporting actress.

S. D. Subbulakshmi married the famous prolific lawyer turned director K. Subramanyam with the consent of his first wife Meenatchi. K. Subramanyam kept both the families together and managed them without any discrimination between them until his last breath. She was blessed with a son Abaswaram Ramji.
